This simple concept hides numerous implications to homeostasis, development and disease, which are here explained. Not only are all basic elements on DAMPs presented, but also comprehensive knowledge on immunology into which DAMPs biology is seamlessly interwoven.

DAMPs bind specific receptors to activate inflammation and start a highly optimized sequence of immune cell recruitment of neutrophils and monocytes to initiate effective tissue repair. When DAMPs are cleared, the recruited leukocytes change from a proinflammatory to a reparative program, a switch that is locally supervised by invariant natural killer T cells. However, not all DAMPs are located intracellularly, they can also be generated by proteases that degrade extracellular matrix and thereby generate DAMPs like hyaluronic acid, 45 fibrinogen, 29 and fibronectin 30 (Table 1). Here, we give an overview of DAMPs that have been associated with nociceptive signaling, with a specific emphasis on HMGB1. This mini-review is focused on plant DAMPs, including the recently discovered Arabidopsis HMGB3, which is the counterpart of the prototypic animal DAMP HMGB1.
